About HPLC System Suitability
System Suitability Testing (SST) ensures that the complete chromatographic system (column, reagents, mobile phase, and instrument) is suitable for the intended analysis.
Parameters
- Theoretical Plates (N): Measures column efficiency. Generally, N > 2000 is required.
- Resolution (Rs): Measures the separation between two peaks. Rs > 2.0 indicates baseline separation.
- Tailing Factor (T): Measures peak symmetry. T should be ≤ 2.0 (USP).
- Capacity Factor (k'): Measures retention. Ideally 2 < k' < 10.
